Mechanical Fingerprint of Senescence in Endothelial Cells

چکیده

Endothelial senescence entails alterations of the healthy cell phenotype, which accumulate over time and contribute to cardiovascular disease. Mechanical aspects regulating adhesion, force generation, response flow senescence-associated drift; however, they remain largely unexplored. Here, we exploit microscopy resolve variations anchoring substrate tractions generated upon aging in nanonewton (nN) range. Senescent endothelial cells display a multifold increase levels basal adhesion generation supported by mature strong focal adhesions. The enhanced mechanical interaction with yields static monolayers that polarize but fail process coordinated shape remodeling reorientation. emerging picture indicates reinforces local may therefore prevent denudation; it compromises ability functionally adapt hemodynamic conditions.

Effects of alpha-mangostin on memory senescence induced by high glucose in human umbilical vein endothelial cells

Objective(s): Hyperglycemia induces cellular senescence in various body cells, such as vascular endothelial cells. Since the vessels are highly distributed in the body and nourish all tissues, vascular damages cause diabetes complications such as kidney failure and visual impairment. Alpha-mangostin is a xanthone found in mangosteen fruit with protective effects in met...

Premature senescence of endothelial cells: Methusaleh's dilemma.

Senescence has been considered a programmed cellular response, parallel to apoptosis, that is turned on when a cell reaches Hayflick's limit. Once cells enter the senescence program, they cease to proliferate and undergo a series of morphological and functional changes. Studies support a central role for Rb protein in controlling this process after it receives senescent signals from the p53 and...

Senescence: Detecting an Evolutionary Fingerprint in Plants

The evolution of a senescent decline in the performance of organisms as they grow old is thought to be an unavoidable aspect of life. A genotype-age interaction in performance is the 'fingerprint' of evolved senescence, which has now for the first time been detected in a plant.

Characterization of cellular senescence mechanisms in human corneal endothelial cells

The human cornea is a tri-laminar structure composed of several cell types with substantial mitotic potential. Age-related changes in the cornea are associated with declining visual acuity and the onset of overt age-related corneal diseases. Corneal transplantation is commonly used to restore vision in patients with damaged or diseased corneas. However, the supply of donor tissue is limited, an...
